Bacteriostatic, bactericidal, and drug resistance studies of ethyl gallate on Mycobacterium tuberculosis.

چکیده
Recent reports show that a compound identified as ethyl gallate (Little et al., 1953) found in Haematoxylon campechianum is specifically inhibitory to Mycobacterium tuberculosis, strain 607 (Johnstone et al., 1953). The antimicrobial activity of gallates is not new information, for several such reports and patents are known (Sabalitschka and Tietz, 1931; Bucher, 1945; and Tollenaar, 1952); but the isolation of ethyl gallate from the plant, the highly specific action against M. tuberculosis, and the demonstration of similar activity of other gallates are, as far as these authors know, the first published information. Waksman and Reilly (1944) after a comparative study of several antibiotics found that they were not only bacteriostatic but also bactericidal. Smith and Waksman (1947) in a study of streptomycin demonstrated it to be both tuberculostatic and tuberculocidal. Since ethyl gallate, as well as two other synthesized esters of gallic acid, namely propyl and butyl gallate, appeared to have similar antibiotic properties, it was felt desirable to determine to what extent this activity was bacteriostatic or bactericidal. In addition, this report contains a study of the ability for M. tuberculosis to become drug resistant and relates to what extent cross resistance occurs among other esters of gallic acid.

A STUDY OF MYCOBACTERIUM TUBERCULOSIS DRUG RESISTANCE IN PULMONARY TUBERCULOSIS
Tuberculosis remains a major public health problem in both developing and developed countries. Drug-resistant tuberculosis is an increasing health problem and serious challenge to tuberculosis (TB) control programs. Information about the susceptibility pattern of Mycobacterium tuberculosis isolates against anti-tuberculosis drugs is an important aspect to TB control. The objectives of the s...
متن کاملEvaluation of Gene Mutations Involved in Drug Resistance in Mycobacterium Tuberculosis Strains Derived from Tuberculosis Patients in Mazandaran, Iran, 2013
Drug resistance (especially multiple drug resistance) in Mycobacterium tuberculosis makes global concerns in treatment and control of tuberculosis. Rapid diagnosis of drug resistant strains of the bacteria has vital importance in the prognosis of the disease. The aim of this study was to identify the mutations responsible for drug resistance in Mycobacterium tuberculosis strains derived from pa...
متن کاملThe combination of sulfamethoxazole, trimethoprim, and isoniazid or rifampin is bactericidal and prevents the emergence of drug resistance in Mycobacterium tuberculosis.
The challenges of developing new drugs to treat tuberculosis (TB) are indicated by the relatively small number of candidates entering clinical trials in the past decade. To overcome these issues, we reexamined two FDA-approved antibacterial drugs, sulfamethoxazole (SMX) and trimethoprim (TMP), for use in TB treatment. SMX and TMP inhibit folic acid biosynthesis and are used in combination to tr...
متن کاملValidation of CoaBC as a Bactericidal Target in the Coenzyme A Pathway of Mycobacterium tuberculosis
Mycobacterium tuberculosis relies on its own ability to biosynthesize coenzyme A to meet the needs of the myriad enzymatic reactions that depend on this cofactor for activity. As such, the essential pantothenate and coenzyme A biosynthesis pathways have attracted attention as targets for tuberculosis drug development. To identify the optimal step for coenzyme A pathway disruption in M. tubercul...

1 Issues in MTBC Drug Susceptibility Testing: Ethambutol BACKGROUND Ethambutol (EMB) is an antituberculosis drug used as part of the first line treatment against Mycobacterium tuberculosis complex (MTBC).1 A primary role of EMB when used in initial combination antituberculosis therapy is to minimize the risk of drug resistance development to companion first line drugs, particularly isoniazid.2 ...
